It is OK for the epoxy to cure before you add more. However, before you put on another coat, you should both sand the old coat, and wash it. Sanding roughs up the surface so you get better epoxy adhesion, and washing removes any amine blush. If your tape is not going to be visible, lots of fill coats are just wasted epoxy. If you want to paint and/or varnish, it, then fill and sand till it's as smooth as you require. Wear a good dust mask - really!
