The glass tape is what ultimately holds the boat together, the fillets just keep the tape from exceeding its minimum bend radius. So as long as the tape doesn't fold tighter than if it was rolled around a nickel, you don't need to add more putty.
Specifically, for that v-joint you mention, just put in enough of a fillet to smooth the transition from one piece to the other, probably 1/4 inch wide.
