No problem. Even if there are color variations after the glass is on, they will be minor and blend in with the variations caused by the woodgrain.

Also, you don't need to sand down to 220 under glass. The glass will be far rougher than that and you'll be filling that weave and sanding that down to 220, so it's a waste of time to do it before glassing. Just make sure it's smooth enough that it doesn't snag the cloth. If you can run your hand over it without getting splinters, your fine (just wipe it down with denatured alcohol after you do that to remove any hand oils).
